چکیده

Mutations in proteins allow functional innovation, but can be critically destabilizing. Recent work shows how chaperonins can rescue innovative mutants, with implications for protein engineering and adaptive evolution.

Functional reconstitution of the nicotinic acetylcholine receptor by CHAPS dialysis depends on the concentrations of salt, lipid, and protein.

The detergent CHAPS was found to be the preferable surfactant for the efficient purification and reconstitution of the Torpedo californica nicotinic acetylcholine receptor (AChR). The main result is that the incorporation of the AChR proteins into lipid vesicles by CHAPS dialysis was strongly dependent on the salt and protein concentrations. As monitored by sucrose gradients, by electron micros...

Systematic analysis of an amidase domain CHAP in 12 Staphylococcus aureus genomes and 44 staphylococcal phage genomes

An alternative treatment for staphylococcal infections caused by antibiotic-resistance strains is to lyse staphylococci with peptidoglycan hydrolases, for example, a cysteine, histidine-dependent amidohydrolase/peptidase (CHAP). Here, CHAPs were analyzed in 12 Staphylococcus aureus genomes and 44 staphylococcal phage genomes. There are 234 putative CHAP-containing proteins and only 64 non-ident...

Solubilization of high affinity G-protein-coupled serotonin1A receptors from bovine hippocampus using pre-micellar CHAPS at low concentration.

The serotonin1A (5-HT1A) receptors are members of a superfamily of seven transmembrane domain receptors that couple to G-proteins. They appear to be involved in various behavioural and cognitive functions. This paper reports an efficient strategy to solubilize 5-HT1A receptors from bovine hippocampal membranes using the zwitterionic detergent CHAPS which is mild and non-denaturing. Since high c...

Nanoscale biofilm modification-method concerning a myoglobin/11-MUA bilayers for bioelectronic device.

We developed surface modification tools for the fabrication of a bioelectronic device which consists of a myoglobin monolayer self-assembled on an 11-MUA layer. To utilize a single protein as the active element, it was necessary to reduce protein aggregation on the protein layer in the nanobio electronic device, which was developed in our previous study and shown to display basic biomemory func...

Arf1p, Chs5p and the ChAPs are required for export of specialized cargo from the Golgi.

In Saccharomyces cerevisiae, the synthesis of chitin is temporally and spatially regulated through the transport of Chs3p (chitin synthase III) to the plasma membrane in the bud neck region. Traffic of Chs3p from the trans-Golgi network (TGN)/early endosome to the plasma membrane requires the function of Chs5p and Chs6p. Chs6p belongs to a family of four proteins that we have named ChAPs for Ch...
